Description

This chemiluminescence immunoassay kit from Assay Genie is intended for the in vitro quantitative determination of Rat Transforming Growth Factor Beta 3 (TGF-beta3) in serum, plasma and other biological fluids. It is a CLIA ELISA Kit based on the Sandwich CLIA principle, using a pre-coated plate pre-coated with an antibody specific to Rat TGF-beta3, a biotinylated detection antibody, and an Avidin-Horseradish Peroxidase (HRP) conjugate added successively to each well. The relative light unit (RLU) value for each well is read on a chemiluminescence immunoassay analyser, and the concentration of TGF-beta3 is calculated by comparing the sample RLU against a standard curve.

The kit reports a sensitivity of 4.69 pg/mL and a detection range of 7.81-500 pg/mL, and it recognises Rat TGF-beta3 in samples with no significant cross-reactivity or interference between Rat TGF-beta3 and analogues. An unopened kit can be stored at 4°C for 1 month, after which components should be stored separately according to the vial labels. Sample preparation guidance covers serum, plasma, cell lysates, tissue homogenates and cell culture supernatant or other biological fluids, with the recommendation to assay stored samples within 7 days at 4°C, otherwise aliquot and store at -20°C (≤1 month) or -80°C (≤3 months) and avoid repeated freeze-thaw cycles.

The assay procedure follows standard sandwich CLIA steps: add 100 µL of standard or sample to each well and incubate for 90 minutes at 37°C; add biotinylated detection antibody working solution and incubate for 1 hour at 37°C; add HRP conjugate working solution and incubate for 30 minutes at 37°C; add substrate mixture and incubate for no more than 5 minutes at 37°C before reading RLU. Results are analysed by plotting a four-parameter logistic curve on log-log graph paper with standard concentration on the x-axis and RLU on the y-axis, and any calculated sample concentration must be multiplied by the dilution factor if samples have been diluted.
