Rat sTNF RI/TNFRSF1A ELISA Kit

A sandwich ELISA kit from Assay Genie designed for the accurate measurement of soluble tumor necrosis factor receptor type 1 (sTNFR1) levels in rat samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit uses a double-antibody sandwich ELISA format with a colorimetric detection method and is suited to research areas including cell death.

The kit has a manufacturer-quoted sensitivity of 9.375 pg/mL and a measuring range of 15.625-1000 pg/mL. Components are stored at 2-8°C for 12 months. The assay protocol runs as follows: equilibrate reagents and TMB substrate to room temperature, then load standards, samples and blanks and incubate the pre-coated plate at 37°C for 90 minutes; add biotin-labelled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; develop with TMB substrate in the dark for 10-20 minutes; add stop solution and read absorbance at 450 nm.

Reported linearity for serum, EDTA plasma and heparin plasma (n=5) at 1:2, 1:4 and 1:8 dilutions is 87-103%, 87-97%, 90-103%; 85-97%, 83-100%, 82-94%; and 80-97%, 81-99%, 83-97% respectively. Recovery ranges and averages are 89-99% (97%) for serum, 85-103% (95%) for EDTA plasma, and 93-101% (97%) for heparin plasma. According to the manufacturer, sTNFR1 is a key receptor in the regulation of the inflammatory response and immune system functioning, with elevated levels associated with rheumatoid arthritis, inflammatory bowel disease and sepsis, supporting research into inflammatory disease pathogenesis and potential therapeutic targets.
