Rat Soluble CD14 ELISA Kit

From Assay Genie, this Sandwich ELISA Kit is designed for the precise measurement of soluble CD14 levels in rat samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Detection is colorimetric, with absorbance read at 450 nm following TMB substrate development.

Quoted performance characteristics include a sensitivity of 0.938 ng/mL and a working range of 1.563–100 ng/mL. Reported linearity (n = 5) at dilutions of 1:2, 1:4 and 1:8 is 85–105%, 86–101% and 86–99% for serum, 85–100%, 84–95% and 80–97% for EDTA plasma, and 88–105%, 91–101% and 80–100% for heparin plasma. Recovery is 94–104% in serum (average 97%), 88–101% in EDTA plasma (average 97%) and 88–98% in heparin plasma (average 91%). The kit is stored at 2–8°C for 12 months.

The assay procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards and samples, incubating the plate at 37°C for 90 minutes for antigen binding, adding biotin-labeled detection antibody with a 60-minute incubation, adding HRP-Streptavidin (SABC) for 30 minutes, developing colour with TMB substrate in the dark for 10–20 minutes, stopping the reaction, and reading absorbance at 450 nm. Soluble CD14 functions as a co-receptor for lipopolysaccharide (LPS) and modulates the inflammatory process, with altered levels associated with inflammatory conditions.
