Description

A colorimetric Sandwich ELISA from Assay Genie is designed for the accurate and precise measurement of SLPI (Secretory Leukocyte Protease Inhibitor) in rat samples, including serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. SLPI is a key protein that plays an important role in immune response and inflammation regulation, protecting mucosal surfaces and exhibiting antimicrobial properties. Abnormal SLPI levels have been linked to inflammatory conditions and respiratory disorders, making the protein a valuable biomarker for studying these conditions and potential therapeutic interventions.

The double-antibody sandwich assay has a reported sensitivity of 0.188 ng/mL and a standard range of 0.313-20 ng/mL. The protocol involves primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, and TMB substrate color development in the dark for 10-20 minutes, with absorbance read at 450 nm immediately after stop solution addition. Uniprot reference: Q9WUQ4. Reported synonyms include SLPI, ALK1, ALP, antileukoproteinase, BLPI, HUSI, HUSI-I, Mucus proteinase inhibitor, Seminal proteinase inhibitor, secretory leukocyte peptidase inhibitor, and Protease inhibitor WAP4.

Linearity (n=5) was reported at 1:2, 1:4, and 1:8 dilutions as 87-102%, 85-102%, and 86-105% for serum; 88-98%, 82-94%, and 85-97% for EDTA plasma; and 90-100%, 87-90%, and 81-97% for heparin plasma. Recovery averaged 95% for serum (range 90-104%), 96% for EDTA plasma (range 85-104%), and 92% for heparin plasma (range 86-98%). The kit should be stored at 2-8°C for 12 months.
