Description

From Assay Genie, this competitive ELISA kit is designed for the quantitative measurement of rat SLCO1A1 (also known as OATP1A1) using a colorimetric detection method. The assay is run in a competitive format on antigen-coated wells and is suitable for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with a reported sensitivity of 0.375 ng/mL and a measurable range of 0.625–40 ng/mL.

SLCO1A1 is a key transporter protein involved in the cellular uptake of endogenous and exogenous compounds such as drugs and toxins, and its dysregulation has been implicated in liver disorders and cancer, highlighting its importance as a potential therapeutic target and biomarker. Reported linearity at 1:2, 1:4, and 1:8 dilutions is 90–98% / 92–102% / 87–99% for serum (n = 10), 87–99% / 84–101% / 82–95% for EDTA plasma (n = 10), and 97–100% / 86–95% / 80–100% for heparin plasma (n = 10). Recovery ranges and averages are 93–103% (98%) for serum (n = 10), 88–104% (94%) for EDTA plasma (n = 10), and 91–104% (99%) for heparin plasma (n = 10).

The procedure involves equilibrating TMB substrate for 30 minutes at room temperature, incubating standard or sample with biotin-labelled antibody at 37°C for 45 minutes, washing the plate, developing with TMB substrate in the dark at 37°C for 10–20 minutes, stopping the reaction, and reading absorbance at 450 nm. The kit should be stored at 2–8°C in a sealed condition and should not be cryopreserved.
