Rat SHBG/Sex Hormone Binding Globulin ELISA Kit

This sandwich ELISA kit from Assay Genie is designed for the precise, quantitative measurement of Sex Hormone Binding Globulin (SHBG) in rat samples. SHBG is a key protein that binds sex hormones such as testosterone and estradiol, regulating their bioavailability and activity; dysregulation of SHBG levels is associated with hormonal imbalances and various health conditions, making it a valuable biomarker for studies in reproductive health, metabolic disorders, and endocrine diseases.

The assay uses a colorimetric sandwich ELISA, double antibody format with a sensitivity of 9.375 pg/mL and a detection range of 15.625-1000 pg/mL. It is reactive with rat SHBG and validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Serum samples are prepared by allowing blood to clot and centrifuging at 1000 × g for 20 minutes; plasma is collected using anticoagulant tubes and centrifuged at 1000 × g for 15 minutes at 2-8°C; cell culture supernatants are centrifuged at 2500 rpm for 5 minutes. Linearity at 1:2 dilution is 88-105% for serum, 83-97% for EDTA plasma, and 80-99% for heparin plasma, with average recovery of 94% for serum, 93% for EDTA plasma, and 96% for heparin plasma.

The kit is supplied with a dismountable ELISA microplate, lyophilized standard, biotin-labeled detection antibody, HRP-streptavidin conjugate, TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, 25X wash buffer, plate sealers, and a technical manual. The assay procedure involves equilibrating reagents to room temperature, primary incubation at 37°C for 90 minutes, detection antibody binding at 37°C for 60 minutes, HRP-streptavidin binding at 37°C for 30 minutes, TMB colour development in the dark for 10-20 minutes, and absorbance measurement at 450 nm after addition of stop solution. Kit components are stored at 2-8°C, with microplate strips and standards also suitable for 12 months at -20°C.
