Rat Serum paraoxonase/lactonase 3 (Pon3) ELISA Kit

Manufactured by Assay Genie, this Sandwich ELISA kit enables quantitative colorimetric measurement of serum paraoxonase/lactonase 3 (Pon3) in rat-derived samples. The assay is intended for use with serum, plasma, tissue homogenates, cell culture supernates, and other biological fluids, and detects the natural and recombinant rat protein (UniProt: Q68FP2), a secreted homodimer reported to hydrolyze certain lactones and organophosphate esters.

Detection is performed on a microplate pre-coated with target-specific antibody, using a biotin-conjugated detection antibody and avidin-HRP, with signal developed by 3,3′,5,5′-Tetramethylbenzidine (TMB) and read at 450 nm. Reported performance includes a detection range of 0.312–20 ng/mL, sensitivity of 0.162 ng/mL, intra-assay CV of 4.9%, and inter-assay CV of 6.2%. Dilutional linearity for rat serum is 94–104% at 1:2, 99–109% at 1:4, 104–113% at 1:8, and 109–121% at 1:16. Spike-and-recovery averages are 85% (range 80–91%) for serum and 87% (range 81–93%) for plasma. The kit additionally includes validated Bradford Reagent for total protein quantification to support sample normalization.

Sample preparation guidance is provided for serum, plasma, urine, cerebrospinal fluid, cell culture supernatant, cell lysates, and tissue homogenates, with repeated freeze–thaw cycles to be avoided.
