Built around the Assay Genie antibody pair raised against rat Semaphorin-3A (UniProt accession Q63548), this quantitative immunoassay detects endogenous, secreted SEMA3A across neuroscience and cardiovascular workflows. The rat target is a class 3 semaphorin that signals through neuropilin-1 and neuropilin-2 co-receptors together with the PLXND1 plexin D1 receptor, enabling studies of axon repulsion, sympathetic nerve patterning, and endothelial repulsion events in vitro and in vivo.

Validated application areas include rat models of spinal cord injury and peripheral nerve crush (regeneration studies), pressure-overload or angiotensin II-induced cardiac remodeling (interstitial fibrosis and perivascular SEMA3A release), unilateral nephrectomy and diabetic nephropathy (glomerular and podocyte injury assessment), and bone remodeling experiments tracking osteoblast and osteoclast crosstalk. Sample preparation typically involves a 1000 x g serum or plasma centrifugation step, with clarified aliquots stored at -80 degrees C and freeze-thaw cycles limited to avoid signal drift.
