P-Selectin (SELP, also known as CD62P or GMP-140) is a calcium-dependent lectin expressed on the surface of activated endothelial cells and platelets, where it mediates leukocyte rolling and adhesion during early inflammatory responses. Researchers use this chemiluminescence immunoassay to quantify circulating SELP levels as a biomarker of vascular activation, thrombosis, and acute inflammation in rat models.
The Assay Genie CLIA Kit reaches lower detection limits than conventional colorimetric ELISA through an acridinium-ester-based luminescent readout measured on a microplate luminometer. Each kit provides a pre-coated 96-well plate along with ready-to-use standards, biotinylated detection antibody, acridinium-ester-labeled streptavidin, and wash buffer sufficient for the full plate. Reconstituted reagents should be stored at 2-8 degrees C and protected from light; standard curves are typically generated from a seven-point dilution series with sample volumes of 50 ul per well.
Validated for serum, plasma (EDTA and heparin), and tissue homogenate supernatants, the assay completes in roughly 3.5 hours and shows minimal cross-reactivity with related rat adhesion molecules such as E-selectin and L-selectin.
