Rat S100A9 ELISA Kit

Reported linearity at 1:2, 1:4 and 1:8 dilutions is 87–102%, 85–101% and 86–103% for serum (n=5), 84–101%, 83–97% and 82–100% for EDTA plasma (n=5), and 80–95%, 81–99% and 88–95% for heparin plasma (n=5). Reported recovery ranges are 90–105% (average 90%) for serum (n=5), 86–104% (average 99%) for EDTA plasma (n=5), and 87–104% (average 97%) for heparin plasma (n=5). Kit components should be stored at 2-8°C for 12 months.

S100A9 (synonym: S100 Calcium Binding Protein A9; Uniprot P50116) is a member of the S100 protein family that plays a role in inflammatory regulation, with elevated levels implicated in rheumatoid arthritis, inflammatory bowel disease and certain cancers. The protocol proceeds by reagent equilibration to room temperature, primary incubation at 37°C for 90 minutes, biotinylated detection antibody binding for 60 minutes at 37°C, HRP-Streptavidin binding for 30 minutes at 37°C, TMB colour development for 10–20 minutes in the dark, and absorbance measurement at 450 nm after stop solution. Serum is clotted and centrifuged at 1000 × g for 20 minutes; plasma is collected with anticoagulant and centrifuged at 1000 × g for 15 minutes at 2–8°C.
