Built around a colorimetric readout read on a standard microplate reader, this Assay Genie ELISA kit targets the rat S100A10 calcium-binding protein, a small member of the S100 family that pairs with Annexin A2 to form the annexin A2 heterotetramer. In rat tissues the protein regulates plasminogen activation, assists in trafficking of serotonin 5-HT1B and TASK-1 potassium channels to the plasma membrane, and supports surface expression of Nav1.8 sodium channels relevant to nociception. Investigators in oncology, neuropharmacology, and vascular biology apply the kit to rat tissue homogenates and cell lysates to quantify S100A10 abundance in models of colorectal and gastric carcinoma, depression-like behavior, and tissue remodeling after vascular injury.

The 96-assay configuration provides microplate strips pre-coated with capture antibody along with lyophilized standards, detection antibody, streptavidin-HRP conjugate, substrate solution, wash buffer, and stop reagent. Reconstituted standards are aliquoted and held refrigerated between uses to limit freeze-thaw loss of immunoreactivity, while unopened kit components are kept refrigerated and protected from light. Standards are run alongside samples, and results are interpolated from a standard curve to express S100A10 concentration in the original homogenate or lysate. The kit is intended for research use only, is not validated for diagnostic procedures, and supports studies examining how altered S100A10 expression contributes to receptor availability at the cell surface, fibrinolysis at the pericellular interface, and disease-associated signaling pathways.
