Each Assay Genie rat S-100 ELISA kit is built around a biotinylated capture and detection antibody pair coupled to streptavidin-HRP, producing a colour change via TMB substrate that is terminated by a sulphuric acid stop solution; a complete microplate run, including incubation and washing steps, takes roughly 3.5 hours from sample loading to plate read. Reproducibility data reported by Assay Genie indicate intra-assay CV below 8% and inter-assay CV below 10% across low, medium and high concentration controls, with spike-and-recovery values falling between 87% and 99% in neat serum, EDTA plasma and heparin plasma, and dilution linearity maintained from 1:2 through 1:16 in each matrix. Specificity testing has shown no significant cross-reactivity with closely related rat S100 analogues, so the measured signal corresponds to the S-100 protein family rather than off-target epitopes.
