Description

From Assay Genie, this Sandwich ELISA is designed for quantitative detection of Retinol-Binding Protein 4 (RBP4) in rat samples using a double-antibody, colorimetric format. RBP4 is a key protein that transports retinol (vitamin A) in the bloodstream and plays a critical role in metabolism and insulin resistance, with dysregulation linked to obesity, diabetes, and cardiovascular disease.

Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported sensitivity is 0.938 ng/mL and detection range is 1.563–100 ng/mL. Linearity for serum (n=5) was 84–99% at 1:2, 88–100% at 1:4, and 83–98% at 1:8; EDTA plasma (n=5) 82–98%, 82–100%, 82–96%; heparin plasma (n=5) 84–100%, 85–99%, 82–100%. Recovery in serum (n=5) was 92–101% (average 94%), EDTA plasma 91–98% (average 97%), heparin plasma 89–104% (average 94%). Components are stored at 2–8°C for 12 months.

The procedure involves equilibrating reagents and TMB substrate, loading standards and samples onto the pre-coated plate for a 90-minute incubation at 37°C, adding biotinylated detection antibody for 60 minutes, binding HRP-Streptavidin (SABC) for 30 minutes, developing colour with TMB substrate in the dark for 10–20 minutes, and reading absorbance at 450 nm after adding stop solution. The associated UniProt entry is P04916; the research area is signal transduction.
