Description

A high-quality sandwich ELISA developed by Assay Genie for the quantitative measurement of Pyruvate dehydrogenase kinase isozyme 1, mitochondrial (PDK1) in rat samples. The kit supports research applications investigating PDK1, a key mitochondrial enzyme that regulates the pyruvate dehydrogenase complex and governs metabolic flux between oxidative phosphorylation and glycolysis. Dysregulation of PDK1 has been linked to metabolic disorders, making it a valuable biomarker for studies of cellular responses to hypoxia and cancer metabolism.

Assay performance is characterised by a sensitivity of 0.081 ng/mL and a detection range of 0.156-10 ng/mL. Intra-assay CV is 4.3% and inter-assay CV is 8.1%. Spike-and-recovery in serum averages 95% (range 89-101%) and in plasma averages 97% (range 91-103%). Linearity has been validated for serum, EDTA plasma, and heparin plasma across dilutions from 1:2 to 1:16. The assay reacts with rat samples and specifically detects natural and recombinant rat PDK1 (UniProt Q63065).

Validated sample types include serum, plasma, tissue homogenates, cell culture supernates, and other biological fluids. PDK1 is a kinase that phosphorylates PDHA1 and PDHA2, inhibiting pyruvate dehydrogenase activity, down-regulating aerobic respiration, and protecting cells against apoptosis under hypoxia and oxidative stress. The protein localises to the mitochondrion matrix, forms homodimers and heterodimers with PDK2, and is a direct HIF-1 target gene. Research area: cancer. For research use only.
