PUMA (BBC3), also known as p53 Upregulated Modulator of Apoptosis, is described as a key regulator of cell death and apoptosis, involved in the response to DNA damage, tumour suppression, and the elimination of damaged cells. The protocol involves equilibrating reagents to room temperature, incubating standards and samples on the pre-coated plate at 37°C for 90 minutes, adding biotin-labelled detection antibody for 60 minutes, applying HRP-Streptavidin (SABC) for 30 minutes, developing colour with TMB substrate for 10–20 minutes in the dark, and measuring absorbance at 450 nm after adding stop solution. The kit is stored at 2-8°C for 12 months. Validated linearity for rat serum ranges from 87-94% to 83-100% across dilutions; EDTA plasma from 89-103% to 82-99%; and heparin plasma from 86-105% to 81-99%. Recovery averages 90% for serum (range 85-103%), 99% for EDTA plasma (range 90-105%), and 94% for heparin plasma (range 88-104%).
