Description

Manufactured by Assay Genie, this Sandwich ELISA uses a double antibody format for the accurate measurement of pulmonary surfactant-associated protein A (SPA) in rat samples. Validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, the kit uses a colorimetric detection method and should be stored at 2-8°C for 12 months.

Performance characteristics include a sensitivity of 0.938 pg/mL and a working range of 1.563-100 pg/mL. Linearity testing showed serum (n=5) recoveries of 85-102%, 82-97% and 80-100% at 1:2, 1:4 and 1:8 dilutions respectively; EDTA plasma (n=5) at 89-100%, 84-90% and 83-98%; and heparin plasma (n=5) at 86-102%, 90-101% and 81-100%. Spike-and-recovery averages were 95% for serum, 96% for EDTA plasma, and 96% for heparin plasma.

SPA is a key protein of pulmonary surfactant that supports proper lung function and prevents respiratory distress, and abnormal SPA levels have been linked with conditions such as acute respiratory distress syndrome (ARDS) and pneumonia. The protocol uses a biotin-labelled detection antibody with HRP-Streptavidin (SABC) and TMB substrate, with primary incubation at 37°C for 90 minutes and absorbance read at 450 nm. The kit is associated with Uniprot P08427.
