P4HB is a multifunctional enzyme that catalyses the formation, breakage and rearrangement of disulfide bonds. At the cell surface it acts as a reductase that cleaves disulfide bonds of proteins attached to the cell, while intracellularly it forms and rearranges disulfide bonds in nascent proteins. At high concentrations it functions as a chaperone that inhibits aggregation of misfolded proteins, and at low concentrations it facilitates aggregation (anti-chaperone activity). It also acts as a structural subunit of prolyl 4-hydroxylase and the microsomal triacylglycerol transfer protein MTTP, and is listed under the Cardiovascular research area.
