Rat PROS1/Protein S ELISA Kit

Manufactured by Assay Genie, this Rat PROS1/Protein S ELISA Kit is specifically designed for the precise measurement of Protein S levels in rat samples. The assay employs a sandwich ELISA format using a double antibody approach, with a reported sensitivity of 0.938 ng/mL and a working range of 1.563–100 ng/mL. It accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and is reactive for rat. The kit should be stored at 2–8°C for 12 months.

Linearity has been validated across serial dilutions of 1:2, 1:4, and 1:8 in serum (93–105%, 91–104%, 86–104%), EDTA plasma (85–101%, 82–87%, 89–98%), and heparin plasma (85–100%, 82–100%, 85–99%). Recovery ranges were 85–104% (average 91%) for serum, 89–99% (average 93%) for EDTA plasma, and 85–103% (average 95%) for heparin plasma.

The assay procedure involves equilibrating reagents and TMB substrate to room temperature, primary incubation of standards and samples at 37°C for 90 minutes, biotin-labeled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) incubation at 37°C for 30 minutes, TMB color development in the dark for 10–20 minutes, and a stop reaction with absorbance measurement at 450 nm. Sample preparation covers serum (clotting, centrifugation at 1000 × g for 20 minutes), plasma (anticoagulant tubes, centrifugation at 1000 × g for 15 minutes at 2–8°C), tissue homogenate (PBS with protease inhibitors), cell culture supernatant (2500 rpm for 5 minutes), and cell lysate workflows.
