Rat Prolyl 4-hydroxylase subunit alpha-1 (P4ha1) ELISA Kit

From Assay Genie, this sandwich ELISA kit is designed for the in vitro quantitative determination of rat Prolyl 4-hydroxylase subunit alpha-1 (P4HA1) in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. The assay is reactive with rat samples and detects both natural and recombinant forms of the target. P4HA1 is an essential subunit of prolyl 4-hydroxylase that catalyses the post-translational formation of 4-hydroxyproline in -Xaa-Pro-Gly- sequences in collagens and other proteins. The protein localises to the endoplasmic reticulum lumen and functions as a heterotetramer of two alpha-1 chains and two beta chains (UniProt P54001).

The colorimetric sandwich ELISA format has a detection range of 0.312–20 ng/mL and a sensitivity of 0.161 ng/mL. Reported precision is an intra-assay CV of 4.8% and an inter-assay CV of 8.9%. Linearity across serial dilutions is 80–123% in serum, 82–110% in EDTA plasma and 80–120% in heparin plasma. Recovery averages 98% in serum (range 92–104%) and 100% in plasma (range 94–106%).

Dysregulation of P4HA1 has been associated with fibrosis, cancer and cardiovascular disorders, supporting its use as a biomarker for research into collagen metabolism and related disease pathways. For research use only.
