Rat PON1/Paraoxonase 1 ELISA Kit

Assay Genie supplies this Sandwich ELISA kit for the quantitative measurement of paraoxonase 1 (PON1) in rat samples. PON1 is an enzyme recognised for its role in detoxifying organophosphates and lipids and for protecting against oxidative stress, and its levels are investigated in the context of cardiovascular disease, diabetes, and neurological disorders.

The assay uses a double-antibody Sandwich ELISA with colorimetric detection. It is intended for rat serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Sensitivity is reported as 0.469 ng/mL with a working range of 0.781-50 ng/mL. Reagents are stored at 2-8°C for 12 months.

Spike-and-recovery testing in serum, EDTA plasma, and heparin plasma produced recovery ranges of 90-104%, 85-104%, and 90-104% with averages of 93%, 92%, and 97% respectively, and linearity between 83% and 101% across 1:2, 1:4, and 1:8 dilutions. The protocol includes reagent equilibration, a 90-minute primary incubation at 37°C, biotinylated detection antibody binding, HRP-streptavidin (SABC) binding, TMB colour development in the dark for 10-20 minutes, and absorbance measurement at 450 nm.
