Description

Assay Genie's Sandwich ELISA kit is designed for the accurate measurement of PMP22 (Peripheral Myelin Protein 22) levels in rat samples, supporting research applications that require sensitivity and specificity. The assay uses a colorimetric detection method in a double antibody sandwich format and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. PMP22 is a vital protein that plays a key role in peripheral nerve myelination and function, and its dysregulation has been linked to peripheral neuropathies such as Charcot-Marie-Tooth disease, making it a valuable biomarker for studying these conditions and researching potential treatment options.

Validated performance includes linearity at 1:2, 1:4, and 1:8 dilutions for serum (85-103%, 85-97%, 85-100%), EDTA plasma (85-103%, 82-101%, 89-100%), and heparin plasma (88-95%, 83-95%, 82-95%). Recovery ranges are 85-103% for serum (average 95%), 87-105% for EDTA plasma (average 96%), and 90-104% for heparin plasma (average 96%). The assay procedure involves equilibrating reagents to room temperature and setting up standard, test, and control wells; primary incubation at 37°C for 90 minutes; biotin-labeled detection antibody binding at 37°C for 60 minutes; HRP-Streptavidin binding at 37°C for 30 minutes; TMB color development in the dark for 10-20 minutes; and a stop reaction with absorbance reading at 450 nm.

Sample preparation guidance includes allowing blood to clot and centrifuging serum at 1000 × g for 20 minutes, collecting plasma using anticoagulant tubes and centrifuging at 1000 × g for 15 minutes at 2-8°C, homogenizing tissue in PBS with protease inhibitors, centrifuging cell culture supernatant at 2500 rpm for 5 minutes, and lysing cells with a buffer containing protease inhibitors. Unopened kit components are stored at 2-8°C for 12 months. The target protein is catalogued under UniProt P25094.
