Description

A sandwich ELISA kit from Assay Genie designed for the quantitative measurement of Plasmin-Antiplasmin Complex (PAP) in rat samples. PAP is described by the manufacturer as a key component of the fibrinolytic system, playing a critical role in clot dissolution and the maintenance of hemostasis. The kit is intended for research into conditions in which PAP dysregulation has been implicated, including cardiovascular disorders, thrombosis, and liver diseases, supporting investigations of the fibrinolytic system and potential therapeutic interventions.

The assay uses a double-antibody sandwich format with colorimetric detection, read at 450 nm after a stop step. It is reactive with rat samples and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Sensitivity is 28.125 pg/mL and the working range is 46.875-3000 pg/mL. The kit should be stored at 2-8°C and remains stable for 12 months under those conditions.

Dilutional linearity for serum (n=5) is 83-101% at 1:2, 80-99% at 1:4, and 85-92% at 1:8; for EDTA plasma (n=5) 84-100%, 81-96%, and 89-102%; and for heparin plasma (n=5) 82-86%, 82-101%, and 83-93%. Spike recovery is 89-104% with an average of 99% in serum, 93-103% with an average of 94% in EDTA plasma, and 86-100% with an average of 98% in heparin plasma. The workflow involves primary incubation at 37°C, biotinylated detection antibody binding, HRP-streptavidin binding, TMB substrate development in the dark, and a stop step before absorbance measurement.
