Rat PKR/EIF2AK2 ELISA Kit

The Assay Genie sandwich ELISA kit is configured as a sandwich ELISA, double antibody assay with colorimetric detection, designed for the precise measurement of PKR (also known as EIF2AK2) in rat samples. It is reactive with rat species and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported sensitivity is 0.188 ng/mL with an assay range of 0.313-20 ng/mL. Components should be stored at 2-8°C for 12 months.

The procedure begins with reagent equilibration to room temperature, followed by loading standards, samples and blanks onto the pre-coated plate for a 90-minute incubation at 37°C. A biotin-labeled detection antibody is added for 60 minutes at 37°C, then HRP-Streptavidin (SABC) for 30 minutes at 37°C. TMB substrate is incubated in the dark for 10-20 minutes, after which stop solution is applied and absorbance read at 450 nm.

Reported linearity for serum (n=5) is 85-104% at 1:2, 85-98% at 1:4, and 89-102% at 1:8 dilutions. EDTA plasma (n=5) shows 82-97%, 82-101%, and 87-100%, while heparin plasma (n=5) gives 80-94%, 90-97%, and 80-98%. Recovery is 98-105% (average 100%) for serum, 85-103% (average 93%) for EDTA plasma, and 87-101% (average 93%) for heparin plasma.
