Rat PKM2 ELISA Kit

This Sandwich ELISA kit from Assay Genie is developed for the precise measurement of pyruvate kinase M2 (PKM2) levels in rat samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. PKM2 is a key enzyme in the glycolytic pathway with a known role in cancer metabolism and tumour growth, supporting research into metabolic reprogramming in cancer cells and potential therapeutic targets.

Using a double antibody sandwich format with colorimetric detection, the kit offers a sensitivity of 46.875 pg/mL and a detection range of 78.125-5000 pg/mL (80IU=5ng). Linearity was demonstrated across dilutions of 1:2, 1:4 and 1:8 for serum (85-96%, 82-97%, 81-99%), EDTA plasma (91-104%, 81-99%, 91-104%) and heparin plasma (82-98%, 82-98%, 83-92%). Recovery ranges are 91-104% for serum (average 95%), 87-98% for EDTA plasma (average 94%) and 88-105% for heparin plasma (average 92%).

The protocol covers reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin incubation at 37°C for 30 minutes, TMB substrate development in the dark for 10-20 minutes, and absorbance reading at 450 nm. The kit is stored at 2-8°C for 12 months and is supplied with a dismountable ELISA microplate, lyophilised standard, biotin-labelled antibody concentrate, HRP-Streptavidin conjugate, TMB substrate, sample, antibody and SABC dilution buffers, stop solution, wash buffer concentrate, plate sealer and technical manual.
