Description

Assay Genie's Rat PACAP ELISA Kit is a Sandwich ELISA, Double Antibody assay designed for the colorimetric quantification of Pituitary adenylate cyclase-activating polypeptide (PACAP), a neuropeptide involved in hormone secretion, cell proliferation, and neuronal survival. The assay targets rat samples and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with detection carried out using HRP-Streptavidin and TMB substrate read at 450 nm.

According to the manufacturer, the kit has a sensitivity of 3.75 pg/mL and a working range of 6.25–400 pg/mL, with reagents stored at 2–8°C for 12 months. Linearity for serum (n=5) was 96–105%, 86–92%, and 91–105% at 1:2, 1:4, and 1:8 dilutions respectively; EDTA plasma (n=5) showed 83–98%, 82–94%, and 85–101%; and heparin plasma (n=5) showed 80–97%, 81–99%, and 80–92%. Recovery averaged 91% for serum (range 85–100%), 94% for EDTA plasma (range 85–99%), and 94% for heparin plasma (range 87–101%).

The procedure involves a 90-minute primary incubation at 37°C, a 60-minute detection antibody binding step, a 30-minute HRP-Streptavidin incubation, and TMB color development in the dark for 10–20 minutes. The kit is intended for the Neuroscience research area and is associated with Uniprot P13589. Synonyms include Adcyap1, PACAP, and ADCYAP1.
