Description

A competitive ELISA kit from Assay Genie designed for the quantitative measurement of Pituitary Adenylate Cyclase Activating Polypeptide 27 (PACAP-27) in rat samples. The assay is reactive with rat and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with a colorimetric detection readout. PACAP-27 is a neuropeptide implicated in neurotransmission, neuroprotection, and neuroendocrine function, and its dysregulation has been linked to various neurological disorders. The kit supports research into hormone secretion, circadian rhythms, and stress responses.

The assay reports a sensitivity of 4.688 pg/mL and a measuring range of 7.813-500 pg/mL. Linearity at 1:2, 1:4, and 1:8 dilutions is 85-102%, 83-99%, and 81-95% for serum (n=10); 85-104%, 83-100%, and 81-95% for EDTA plasma (n=10); and 87-105%, 86-95%, and 83-100% for heparin plasma (n=10). Recovery ranges are 85-105% (average 92%) for serum, 85-99% (average 92%) for EDTA plasma, and 90-103% (average 99%) for heparin plasma. The protocol involves adding 50 µL of standard or sample together with 50 µL of biotin-labeled antibody per well, incubating at 37°C for 45 minutes, washing, TMB colour development in the dark at 37°C for 10-20 minutes, adding stop solution, and reading absorbance at 450 nm. Kits should be stored sealed at 2-8°C and should not be cryopreserved.
