Description

From Assay Genie, this Rat Phospho-TAU/pTAU ELISA Kit is designed for the precise measurement of phosphorylated tau protein levels in rat samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, cerebrospinal fluid (CSF), and other liquid samples. Phosphorylated tau protein plays a role in the pathogenesis of neurodegenerative disorders, particularly Alzheimer's disease, and quantifying pTau levels supports research into disease progression, biomarker discovery, and potential therapeutic interventions.

The kit uses a sandwich ELISA, double antibody format with colorimetric detection. It has a sensitivity of 9.375 pg/mL and a measurable range of 15.625-1000 pg/mL. Storage is at 2-8°C for 12 months. Recovery values reported for serum (n=5) range from 85-102% with an average of 96%, EDTA plasma (n=5) from 88-100% with an average of 97%, and heparin plasma (n=5) from 88-105% with an average of 95%. Linearity data for serum (n=5) spans 87-96% at 1:2, 83-101% at 1:4, and 89-99% at 1:8; EDTA plasma (n=5) shows 83-99%, 80-100%, and 82-101% at the same dilutions; heparin plasma (n=5) shows 80-98%, 80-94%, and 83-95%.

The assay procedure involves equilibrating reagents and TMB substrate to room temperature, incubating the plate at 37°C for 90 minutes to allow antigen binding, adding biotin-labeled detection antibody with a 60-minute incubation at 37°C, adding HRP-Streptavidin (SABC) for 30 minutes at 37°C, developing colour with TMB substrate in the dark for 10–20 minutes, and reading absorbance at 450 nm after adding stop solution. Sample preparation protocols are provided for serum, plasma, tissue homogenate, cell culture supernatant, and cell lysate. The kit has been cited in studies by Braissant et al. investigating liver dysfunction and early Alzheimer's pathology in adult rat models of chronic liver disease.
