Rat Peptidyl-prolyl cis-trans isomerase A (Ppia) ELISA Kit

This Sandwich ELISA kit from Assay Genie enables quantitative detection of Rat Peptidyl-prolyl cis-trans isomerase A (PPIA), a protein also known as Cyclophilin A, Cyclosporin A-binding protein, Rotamase A, p1B15, p31 and PPIase A. The assay is reactive with rat samples and is validated for serum, plasma, tissue homogenates, cell culture supernatants and other biological fluids.

Performance specifications include a detection range of 0.156–10 ng/mL, a sensitivity of 0.087 ng/mL, an intra-assay CV of 4.6% and an inter-assay CV of 6.6%, using a colorimetric detection principle. The antibody pair specifically recognises natural and recombinant rat PPIA. Linear dilution performance has been validated across 1:2, 1:4, 1:8 and 1:16 dilutions, with serum values of 97–108%, 107–117%, 103–111% and 90–101%, EDTA plasma values of 99–108%, 85%, 103–112% and 90–100%, and heparin plasma values of 108–117%, 87–98%, 104–114% and 93–102%. Average recovery is 87% for serum (range 81–93%) and 89% for plasma (range 83–95%).

PPIA is a peptidyl-prolyl cis-trans isomerase that accelerates protein folding by catalysing the cis–trans isomerization of proline imidic peptide bonds in oligopeptides. The protein is localised to the cytoplasm and is also secreted; secretion in vascular smooth muscle occurs in response to oxidative stress via a vesicular pathway involving actin remodelling and myosin II activation and mediates ERK1/2 activation. Reagents are stored at –20 °C with selected buffer components held at 4 °C, and the kit is intended for research use only.
