Description

A sandwich ELISA, double antibody assay from Assay Genie developed for the precise measurement of PECAM1 (CD31) levels in rat samples using a colorimetric detection method. The kit is reactive with rat and is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. PECAM1 (CD31) is a key protein in endothelial cell biology, playing a critical role in cell adhesion, migration, and angiogenesis, and its dysregulation has been linked to inflammation, atherosclerosis, and cancer, making it a valuable biomarker for vascular biology research.

Performance specifications include a sensitivity of 0.938 ng/mL and an assay range of 1.563–100 ng/mL. Linearity testing (n=5 per sample type) returned 88–103%, 87–101% and 87–94% for serum at 1:2, 1:4 and 1:8 dilutions respectively; 84–101%, 84–96% and 83–99% for EDTA plasma; and 82–97%, 83–89% and 82–96% for heparin plasma. Recovery testing showed average values of 93% for serum (range 89–99%), 91% for EDTA plasma (range 85–105%) and 95% for heparin plasma (range 86–100%).

The protocol proceeds through reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, TMB substrate colour development in the dark for 10–20 minutes, and stop solution addition with absorbance reading at 450 nm. Unopened kit components are stored at 2–8°C for 12 months.
