Description

From Assay Genie, this Sandwich ELISA kit is designed for the accurate measurement of pyruvate dehydrogenase E1 component subunit alpha (PDHA1) levels in rat samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. PDHA1 is a key enzyme in the pyruvate dehydrogenase complex, playing a crucial role in the conversion of pyruvate to acetyl-CoA in the citric acid cycle. Dysregulation of PDHA1 has been linked to metabolic disorders, neurodegenerative diseases, and cancer, making it a valuable biomarker for studying these conditions and discovering potential therapeutic interventions.

The assay uses a double antibody sandwich format with colorimetric detection. It offers a sensitivity of 0.188 ng/mL and a range of 0.313-20 ng/mL. Linearity for serum (n=5) is reported as 86-98% at 1:2, 85-104% at 1:4, and 89-104% at 1:8; for EDTA plasma (n=5) as 82-95%, 85-100%, and 83-99%; and for heparin plasma (n=5) as 86-98%, 82-99%, and 80-100%. Recovery ranges are 87-99% (average 95%) for serum, 89-104% (average 96%) for EDTA plasma, and 87-103% (average 98%) for heparin plasma. The kit should be stored at 2-8°C for 12 months.

The protocol involves equilibrating reagents and TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, TMB color development in the dark for 10-20 minutes, and reading absorbance at 450 nm after adding stop solution. The kit supports metabolism research, enabling the investigation of metabolic pathways and the development of novel treatment strategies.
