Rat PDGFRL ELISA Kit

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative measurement of PDGFRL in rat samples, supporting research into the protein's role in cell growth, proliferation and differentiation. Using a double-antibody sandwich format with colorimetric detection, the kit delivers a sensitivity of 46.875 pg/mL and a working range of 78.125–5000 pg/mL. It is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types, with recommended storage at 2–8°C for 12 months.

The assay is carried out on a pre-coated microplate. After equilibrating reagents and TMB substrate to room temperature, standards, samples and blanks are loaded and incubated at 37°C for 90 minutes. A biotin-labelled detection antibody is then added for a 37°C, 60-minute incubation, followed by HRP-Streptavidin (SABC) at 37°C for 30 minutes. TMB substrate is added and developed in the dark for 10–20 minutes before the stop solution is applied and absorbance is read at 450 nm. Sample preparation calls for clotting and centrifugation of serum at 1000 × g for 20 minutes, plasma collection in anticoagulant tubes with centrifugation at 1000 × g for 15 minutes at 2–8°C, and clarification of cell culture supernatant at 2500 rpm for 5 minutes.

Linearity in serum (n=5) was 87–101% at 1:2, 85–105% at 1:4 and 85–92% at 1:8; in EDTA plasma 92–100%, 85–99% and 82–98%; and in heparin plasma 82–96%, 84–98% and 84–95%. Corresponding recovery ranges and averages were 89–105% (100%) for serum, 89–98% (94%) for EDTA plasma and 87–103% (95%) for heparin plasma.
