Rat PDGF-AB/Platelet-Derived Growth Factor AB ELISA Kit

A Sandwich ELISA, Double Antibody kit from Assay Genie designed for the quantitative detection of Platelet-Derived Growth Factor AB (PDGF-AB) in rat samples. The colorimetric assay is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and is stored at 2-8°C for 12 months.

Sensitivity is 4.688 pg/mL, with a working range of 7.813-500 pg/mL. Linearity in serum (n = 5) is reported as 86-104% at 1:2, 84-98% at 1:4 and 86-99% at 1:8; EDTA plasma (n = 5) gives 91-103%, 87-101% and 80-100% at the same dilutions; heparin plasma (n = 5) gives 95-105%, 82-97% and 83-97%. Recovery ranges are 87-99% (serum, average 93%), 88-97% (EDTA plasma, average 92%) and 90-103% (heparin plasma, average 96%).

The protocol involves equilibrating reagents and TMB substrate to room temperature, followed by a 90-minute primary incubation at 37°C, a 60-minute incubation with the biotin-labeled detection antibody, a 30-minute HRP-Streptavidin (SABC) step, and 10-20 minutes of TMB substrate development in the dark. Absorbance is read at 450 nm after addition of stop solution.
