Description

From Assay Genie, this Rat PDE4B ELISA Kit is a sandwich ELISA using a double-antibody format for the quantitative measurement of cAMP-specific 3',5'-cyclic phosphodiesterase 4B (PDE4B) in rat samples. PDE4B is an enzyme that regulates intracellular levels of cAMP, a key second messenger involved in many cellular processes, and dysregulation of PDE4B has been implicated in inflammatory, neurological and cancer-related research contexts.

The kit accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and is stored at 2-8°C for 12 months. It delivers a sensitivity of 46.875 pg/mL across a working range of 78.125-5000 pg/mL, with absorbance read at 450 nm following TMB-based color development. Reported linearity across serial dilutions spans 86-102% for serum, 89-99% for EDTA plasma, and 81-98% for heparin plasma, while spike-and-recovery averages 100% in serum, 94% in EDTA plasma, and 92% in heparin plasma.

The workflow involves equilibrating reagents and a pre-coated plate, a primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding for 60 minutes, HRP-streptavidin binding for 30 minutes, and TMB colour development in the dark for 10-20 minutes, before stop solution is added and absorbance is measured immediately at 450 nm.
