Rat PD-L1/CD274 ELISA Kit

Designed for the precise measurement of programmed death-ligand 1 (PD-L1) in rat samples, the Assay Genie Rat PD-L1/CD274 ELISA Kit uses a sandwich ELISA, double antibody format compatible with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. PD-L1, also known as CD274, is a key immune checkpoint protein involved in regulating immune responses and maintaining immune tolerance, with abnormal expression linked to cancer and autoimmune disorders.

The colorimetric assay offers a sensitivity of 46.875 pg/mL and a measurement range of 78.125-5000 pg/mL. Linearity across dilution factors 1:2, 1:4, and 1:8 was 86-102%, 87-103%, and 92-102% for serum (n=5); 82-98%, 93-101%, and 92-99% for EDTA plasma (n=5); and 86-99%, 85-101%, and 82-99% for heparin plasma (n=5). Recovery ranged from 85-102% (average 96%) in serum, 88-99% (average 97%) in EDTA plasma, and 88-104% (average 98%) in heparin plasma. Kits are stored at 2-8°C for 12 months.

The protocol involves equilibrating reagents to room temperature, incubating standards and samples at 37°C for 90 minutes, adding biotin-labeled detection antibody for 60 minutes, binding HRP-Streptavidin for 30 minutes, developing with TMB substrate in the dark for 10-20 minutes, and reading absorbance at 450 nm after adding stop solution.
