Rat PARP1 ELISA Kit

A Sandwich ELISA kit from Assay Genie designed for the quantitative measurement of Poly(ADP-ribose) polymerase 1 (PARP1) in rat samples. The assay uses a colorimetric, double-antibody format and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types.

Reported sensitivity is 46.875 pg/mL with an assay range of 78.125-5000 pg/mL. The kit is stored at 2-8°C for 12 months. Linearity in serum (n = 5) ranges 87-104% at 1:2, 83-98% at 1:4 and 90-99% at 1:8; EDTA plasma (n = 5) ranges 87-102%, 92-98% and 82-97% at the same dilutions; heparin plasma (n = 5) ranges 88-103%, 84-98% and 82-92%. Recovery averages are 97% (serum, n = 5, range 89-102%), 100% (EDTA plasma, n = 5, range 91-104%) and 96% (heparin plasma, n = 5, range 86-104%).

The procedure involves equilibrating reagents and TMB substrate, loading standards and samples for a 90-minute primary incubation at 37°C, adding a biotin-labeled detection antibody for 60 minutes at 37°C, binding HRP-Streptavidin (SABC) for 30 minutes at 37°C, developing with TMB substrate for 10-20 minutes in the dark, stopping the reaction and reading absorbance at 450 nm.
