Rat OTC/Ornithine Carbamoyltransferase ELISA Kit

A sandwich ELISA (double antibody) from Assay Genie is designed for the quantitative measurement of ornithine carbamoyltransferase (OTC), a mitochondrial enzyme involved in the urea cycle with a role in ammonia detoxification and amino acid metabolism. The kit is reactive with rat samples and is labelled for research use only.

Manufacturer-stated performance figures include a sensitivity of 0.938 mIU/mL and an assay range of 1.563–100 mIU/mL. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Storage is stated as 2-8°C for 12 months. Reported linearity across 1:2, 1:4 and 1:8 dilutions (n=5) falls within 81–104% for serum, EDTA plasma and heparin plasma, with average spike-and-recovery values of 96% in serum, 93% in EDTA plasma and 95% in heparin plasma.

The protocol involves a 90 minute primary incubation at 37°C, a 60 minute biotin-labelled detection antibody step, a 30 minute HRP-Streptavidin (SABC) incubation, TMB colour development for 10–20 minutes in the dark, and absorbance measurement at 450 nm after addition of stop solution. Capture antibody is pre-coated onto the microplate, and supplied reagents include a lyophilised standard, 100X biotin-labelled antibody, 100X HRP-Streptavidin conjugate, TMB substrate, sample/antibody/SABC dilution buffers, stop solution, 25X wash buffer and plate sealers.
