Description

A competitive ELISA from Assay Genie designed for the precise measurement of oxytocin (OT) levels in rat serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. The assay uses a colorimetric detection method with the microplate coated with antigen. Oxytocin is a key hormone involved in reproductive functions, social bonding, and emotional regulation, and dysregulation of its levels has been linked to conditions such as autism, anxiety, and depression, making this kit a useful tool for rat model research in these areas.

Performance specifications include a sensitivity of 9.375 pg/mL and an assay range of 15.625–1000 pg/mL. Linearity in serum (n=10) is 91–104% at 1:2, 87–99% at 1:4 and 85–105% at 1:8 dilution; in EDTA plasma (n=10) 82–97%, 85–98% and 83–98%; and in heparin plasma (n=10) 88–99%, 81–96% and 89–97%. Average recovery is 98% for serum, 94% for EDTA plasma and 98% for heparin plasma. Kit components should be stored sealed at 2–8°C and must not be cryopreserved.

The protocol involves equilibrating TMB substrate for 30 minutes at room temperature, adding 50 µL of standard or sample followed by 50 µL of biotin-labelled antibody per well, incubation at 37°C for 45 minutes, washing the plate five times, TMB colour development in the dark at 37°C for 10–20 minutes, addition of stop solution (blue to yellow colour change), and absorbance measurement at 450 nm on a microplate reader.
