Rat Orexin A ELISA Kit

Orexin-A, also known as hypocretin-1, is a neuropeptide that plays a crucial role in regulating wakefulness and sleep, and dysfunction of the orexin system has been linked to sleep disorders including narcolepsy and insomnia.

The assay has a sensitivity of 37.5 pg/mL and operates across a measurement range of 62.5-4000 pg/mL. It is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Storage is recommended at 2-8°C (sealed), with no cryopreservation. Recovery was 86-99% (average 93%) for serum, 87-100% (average 93%) for EDTA plasma, and 91-105% (average 99%) for heparin plasma.

The procedure involves equilibrating TMB substrate for 30 minutes at room temperature, incubating standard or sample with biotin-labeled antibody at 37°C for 45 minutes, washing the plate five times with wash buffer, adding TMB substrate for 10-20 minutes of colour development in the dark, stopping the reaction (colour changes from blue to yellow), and reading absorbance at 450 nm.
