Quantitative measurement of the N-Methyl-D-Aspartate receptor subunit NR1 (also called GluN1) in rat specimens is supported by this immunoassay kit from Assay Genie. NR1 is the obligatory subunit of native NMDA receptors, calcium-permeable ligand-gated ion channels that mediate excitatory glutamatergic neurotransmission throughout the central nervous system and play established roles in synaptic plasticity, learning, memory consolidation, and excitotoxic injury pathways.

Configured in a 96-assay format with antibody pre-coated microplate wells, the kit includes recombinant standards, biotinylated detection antibody, streptavidin-HRP conjugate, chromogenic substrate solution, stop reagent, wash buffer concentrate, and plate sealers sufficient for a full plate run. Researchers load standards and diluted specimens into the coated wells, allow target capture, wash, add the biotinylated detector, incubate, wash again, add the HRP conjugate, develop with substrate, stop the reaction, and read absorbance on a standard plate reader. Signal intensity scales with the concentration of NR1 captured from each specimen, allowing interpolation against a freshly prepared standard curve.

For pre-analytical handling, the kit's validated liquid specimen types should be collected using standard phlebotomy tubes, processed to separate the cell-free fraction, aliquoted into clean storage tubes, and held under refrigerated conditions for short-term work or stored frozen for longer-term retention. Repeated freeze-thaw cycles degrade the analyte and should be minimized. Reagents should be equilibrated to room temperature before use, and light-sensitive components must be shielded from direct exposure during storage and incubation steps.

The kit is intended for in vitro research use only and is suitable for neuroscience and pharmacology laboratories investigating NMDA receptor expression, ischemic brain injury models, neurodegenerative disease research, antiepileptic compound screening, and behavioural neuroscience studies where peripheral or circulating NR1 measurements are of interest. Assay Genie supplies a detailed protocol insert and provides technical support for sample dilution optimization and plate layout decisions across multi-plate campaigns.
