Description

From Assay Genie, this Sandwich ELISA kit uses a double-antibody format for the colorimetric quantification of NKAP (NF-κB Activating Protein) in rat samples. The assay is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and is intended for signal transduction research.

Reported performance includes a sensitivity of 46.875 pg/mL and a working range of 78.125–5000 pg/mL. At 1:2, 1:4, and 1:8 dilutions, serum linearity ranges were 85–103%, 86–102%, and 90–99%; EDTA plasma were 84–101%, 86–99%, and 82–91%; and heparin plasma were 81–92%, 83–99%, and 92–100% (n = 5 per matrix). Spike-and-recovery averages are 93% (serum, range 87–97%), 94% (EDTA plasma, range 85–103%), and 100% (heparin plasma, range 96–105%), with n = 5 per matrix. Unopened components are stored at 2–8°C for 12 months.

The procedure uses a biotin-labelled detection antibody followed by HRP-Streptavidin (SABC), with TMB substrate development and absorbance read at 450 nm. Plates are incubated at 37°C for 90 minutes for primary antigen binding, 60 minutes with detection antibody, and 30 minutes with SABC, before colour development in the dark for 10–20 minutes.
