NAMPT catalyzes the condensation of nicotinamide with 5-phosphoribosyl-1-pyrophosphate to yield nicotinamide mononucleotide, an intermediate in NAD biosynthesis, and is described as the rate-limiting component of the mammalian NAD biosynthesis pathway. The secreted form is reported to behave both as a cytokine with immunomodulating properties and as an adipokine with anti-diabetic properties, and the protein has been linked to modulation of circadian clock function via SIRT1-mediated effects on CLOCK-ARNTL/BMAL1.
