Description

Assay Genie's colorimetric ELISA kit is designed for the accurate and sensitive detection of Neurokinin 1 Receptor (NK1R) levels in rat samples. The Neurokinin 1 Receptor is a protein involved in various physiological and pathological processes in the central nervous system, playing a key role in pain transmission, stress responses, and behavioral regulation, making it a valuable biomarker for studying neurological disorders and potential therapeutic interventions. The kit is configured as a Sandwich ELISA using a double antibody method.

It is intended for use with rat samples and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit has a sensitivity of 75 pg/mL and an assay range of 125-8000 pg/mL. It should be stored at 2-8°C for 12 months. The assay procedure involves equilibrating reagents and TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, TMB substrate color development in the dark for 10-20 minutes, and reading absorbance at 450 nm immediately after adding the stop solution.

Reported linearity values are: serum (n=5) 89-100% at 1:2, 81-86% at 1:4, 81-93% at 1:8; EDTA plasma (n=5) 84-98% at 1:2, 90-105% at 1:4, 89-104% at 1:8; heparin plasma (n=5) 85-103% at 1:2, 83-92% at 1:4, 91-100% at 1:8. Reported recovery values are: serum (n=5) 90-103% with average 92%; EDTA plasma (n=5) 86-105% with average 96%; heparin plasma (n=5) 85-101% with average 91%. The target is also known by the synonyms TACR1, SPR, TAC1R, NK-1R, NK1R, neurokinin 1 receptor, NKIR, substance-P receptor, tachykinin 1 receptor, substance P receptor, tachykinin receptor 1, neurokinin-1 receptor, and NK-1.
