Description

This Sandwich ELISA kit from Assay Genie is designed for the quantitative detection of Neurochondrin (NCDN), also referred to as Norbin, in rat-derived samples including serum, plasma, and cell culture supernatants. The assay uses a sandwich-based, colorimetric detection method and is supplied for research use only, with a stated shelf life of 12 months under appropriate storage conditions.

Neurochondrin is implicated in neural development and function, contributing to synaptic transmission and plasticity, and its dysregulation has been linked to neurological disorders such as epilepsy and schizophrenia, making it a valuable biomarker for investigating these conditions. The assay covers a working range of 0.156–10 ng/mL. Reported recovery rates from spiked samples were 80–102% (average 91%) in serum, 81–100% (average 90%) in EDTA plasma, and 80–89% (average 84%) in heparin plasma. Linearity across 1:2 to 1:16 dilutions was demonstrated for serum (82–96%, 83–98%, 81–99%, 93–101%), EDTA plasma (88–101%, 86–95%, 90–102%, 78–93%), and heparin plasma (80–91%, 82–90%, 95–104%, 79–95%). Intra-assay precision is reported as CV <10% and inter-assay precision as CV <12%, with kit stability showing a loss of activity of less than 5% within the stated expiration date.

The protocol involves preparing reagents and standards, adding 100 µL of standard or sample per well and incubating for 2 hours at 37 °C, followed by addition of 100 µL of Detection Reagent A (1 hour at 37 °C), washing, addition of 100 µL of Detection Reagent B (1 hour at 37 °C), further washing, incubation with 90 µL of Substrate Solution for 15–25 minutes at 37 °C, and addition of 50 µL of Stop Solution before reading at 450 nm.
