Description

Designed for the precise measurement of N-terminal propeptide of Collagen alpha-1 (II) chain (PIINP) levels in rat samples, this sandwich ELISA kit from Assay Genie uses a double-antibody format. PIINP is a recognised marker of collagen synthesis, particularly for the type II collagen found in cartilage, and is regarded as a valuable target for research into cartilage maintenance and repair, as well as conditions such as osteoarthritis, rheumatoid arthritis and other musculoskeletal disorders.

Reactivity is rat, and accepted sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Linearity across serum, EDTA plasma and heparin plasma at 1:2, 1:4 and 1:8 dilutions is reported as 97–101% / 86–100% / 82–97% (serum), 90–105% / 83–97% / 82–100% (EDTA plasma) and 87–102% / 84–98% / 83–99% (heparin plasma), with n=5 for each. Recovery averages are 92% (serum), 97% (EDTA plasma) and 98% (heparin plasma), with corresponding ranges of 86–104%, 87–104% and 91–101%. Storage is at 2–8°C for 12 months.

The workflow follows a standard sandwich ELISA sequence: equilibrate reagents and TMB substrate to room temperature and load standards, samples and controls onto the pre-coated plate; incubate at 37°C for 90 minutes; add biotin-labelled detection antibody for 60 minutes at 37°C; add HRP-Streptavidin (SABC) for 30 minutes at 37°C; develop with TMB substrate in the dark for 10–20 minutes; add stop solution and read absorbance at 450 nm immediately.
