Rat MMP 9 ELISA Kit

Assay Genie supplies a sandwich-format, double-antibody ELISA for the quantitative measurement of MMP-9 in rat samples, using a colorimetric detection method. The assay has a stated sensitivity of 46.875 pg/mL and a working range of 78.125–5000 pg/mL.

Validated sample matrices include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reagents are stored at 2–8°C and remain stable for 12 months. Sample preparation guidance covers centrifugation of clotted blood for serum (1000 × g, 20 minutes) and anticoagulated blood for plasma (1000 × g, 15 minutes at 2–8°C), as well as handling of tissue homogenates, cell culture supernatants and cell lysates. Reported spike-and-recovery values average 98% in serum (range 90–105%, n=5), 93% in EDTA plasma (86–104%) and 95% in heparin plasma (87–100%).

The procedure involves equilibrating reagents to room temperature, a 37°C primary incubation for 90 minutes, detection antibody binding for 60 minutes, HRP-streptavidin incubation for 30 minutes, and TMB substrate development in the dark for 10–20 minutes; absorbance is read at 450 nm after addition of stop solution. MMP-9 (also referred to as Matrix Metalloproteinase 9, Gelatinase B, or CLG4B) is an enzyme involved in extracellular matrix remodelling, cell migration and tissue repair, with dysregulation linked to cancer, inflammation and cardiovascular disease research.
