Description

Manufactured by Assay Genie, this sandwich ELISA kit (double-antibody, colorimetric detection) is designed for the accurate quantification of MIP-3 beta, also known as CCL19, a chemokine involved in the recruitment of immune cells to inflammatory sites and in the regulation of immune responses and inflammation. The kit is intended for use with rat-derived samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, supporting a variety of research applications relating to autoimmune disorders, allergies, and inflammatory conditions.

According to the product specification, sensitivity is 9.375 pg/mL and the assay range is 15.625–1000 pg/mL. Reported linearity across 1:2, 1:4 and 1:8 dilutions (n = 5) is 87–103% / 94–101% / 85–101% for serum, 84–101% / 83–94% / 85–96% for EDTA plasma, and 83–100% / 81–99% / 83–100% for heparin plasma. Reported recovery ranges are 87–99% (average 94%) for serum, 84–99% (average 92%) for EDTA plasma, and 86–105% (average 100%) for heparin plasma. The kit should be stored at 2–8°C for 12 months.

The assay procedure follows a six-step sandwich ELISA workflow: equilibrate reagents and TMB substrate to room temperature and set up standard, test and zero wells on the pre-coated plate; incubate standards, samples and blanks at 37°C for 90 minutes for antigen binding; add biotin-labelled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; develop colour with TMB substrate in the dark for 10–20 minutes; and stop the reaction and read absorbance at 450 nm. Sample preparation guidance includes clotting and centrifugation at 1000 × g for 20 minutes for serum, anticoagulant collection with centrifugation at 1000 × g for 15 minutes at 2–8°C for plasma, homogenisation in PBS with protease inhibitors for tissue, centrifugation at 2500 rpm for 5 minutes for cell culture supernatant, and lysis with protease inhibitors for cell lysates.
