Description

Assay Genie supplies a sandwich-format ELISA kit for the quantitative measurement of Melanocortin receptor 4 (Mc4r) in rat samples, using a colorimetric detection method. The assay is intended for research use only and is validated for serum, plasma, tissue homogenates, cell culture supernates and other biological fluids, with reactivity demonstrated against rat material. Specificity covers natural and recombinant rat Melanocortin receptor 4.

The reported detection range is 0.312-20ng/mL with a sensitivity of 0.103ng/mL. Intra-assay precision is 4.1% CV and inter-assay precision is 8.3% CV. Linearity in serum (N=5) is reported as 88-97% at 1:2, 95-106% at 1:4, 106-115% at 1:8 and 95-106% at 1:16; EDTA plasma (N=5) as 83-91% at 1:2, 107-116% at 1:4, 118-128% at 1:8 and 91-101% at 1:16; heparin plasma (N=5) as 88-98% at 1:2, 109-118% at 1:4, 101-111% at 1:8 and 100-109% at 1:16. Recovery averages 101% for serum (range 95-107%) and 103% for plasma (range 97-109%).

Functionally, Mc4r is described as a receptor specific to the heptapeptide core common to adrenocorticotropic hormone and alpha-, beta-, and gamma-MSH, playing a central role in energy homeostasis and somatic growth and signalling through G proteins that stimulate adenylate cyclase (cAMP). The receptor is localised to the cell membrane as a multi-pass membrane protein, with a reported molecular weight of 36,887 Da and UniProt accession P70596. The kit is positioned within cardiovascular research.
