Description

Designed by Assay Genie for the accurate detection of lanosterol synthase levels in rat samples, this Sandwich ELISA kit uses a double-antibody format with a colorimetric readout. Lanosterol synthase is a key enzyme in the cholesterol biosynthesis pathway, converting 2,3-oxidosqualene to lanosterol, and its dysregulation has been implicated in various metabolic disorders, making it a valuable target for studying cholesterol metabolism and potential therapeutic interventions.

The kit is reactive with rat samples and has been validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported sensitivity is 0.188 nmol/mL with a working range of 0.313-20 nmol/mL. Linearity testing across serum, EDTA plasma, and heparin plasma (n = 5 per matrix) yielded dilution recoveries broadly spanning 82-105% across 1:2, 1:4, and 1:8 dilutions, while spiked recovery averaged 96% in serum, 97% in EDTA plasma, and 96% in heparin plasma. The assay uses an HRP-streptavidin detection system with TMB substrate, read at 450 nm after a stop step, and components are intended to be stored at 2-8°C (with the microplate strips and lyophilized standards also suitable at -20°C for longer-term storage) for up to 12 months.

Reagents supplied include a dismountable pre-coated microplate, lyophilized standard, 100X biotin-labeled detection antibody, 100X HRP-streptavidin conjugate (SABC), TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, 25X wash buffer, plate sealers, and a technical manual. The associated UniProt entry is P48450, the product is classified under the Metabolism research area, and listed synonyms include Lss, Lanosterol synthase, LSS, 2,3-epoxysqualene--lanosterol cyclase, Oxidosqualene--lanosterol cyclase, and OSC.
