Description

From Assay Genie, this Sandwich ELISA uses a double-antibody format for the quantitative detection of rat L-lactate dehydrogenase A chain (LDHA). The assay is a colorimetric method with a sensitivity of 0.469 ng/mL and a working range of 0.781–50 ng/mL, and it is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reagents are intended to be stored at 2–8°C for 12 months.

Linearity has been assessed across serum, EDTA plasma, and heparin plasma samples (n = 5) at dilution factors of 1:2, 1:4, and 1:8. Serum showed recoveries between 93–102%, 83–96%, and 81–93% across those dilutions; EDTA plasma between 86–104%, 82–100%, and 81–100%; and heparin plasma between 86–98%, 82–96%, and 85–97%. Spike-and-recovery testing (n = 5) returned averages of 98% for serum (range 91–102%), 94% for EDTA plasma (range 85–98%), and 99% for heparin plasma (range 89–103%).

LDHA catalyses the conversion of pyruvate to lactate during anaerobic metabolism, and the kit is positioned for research in metabolism, cancer, and cardiovascular disease. The workflow involves equilibrating reagents to room temperature, an initial 90-minute incubation at 37°C for antigen binding, a 60-minute biotin-labelled detection antibody step, 30 minutes with HRP-Streptavidin, 10–20 minutes of TMB substrate development in the dark, and absorbance reading at 450 nm after stop solution addition.
