Description

Assay Genie's Rat Laminin ELISA Kit is a sandwich ELISA developed for the quantitative measurement of laminin in rat samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay uses a colorimetric detection method and a double-antibody sandwich format, with a stated sensitivity of 75 pg/mL and a working range of 125-8000 pg/mL. Laminin is a key component of the extracellular matrix involved in cell adhesion, migration and differentiation, and altered laminin expression has been associated with cancer, muscular dystrophy and kidney disorders.

The protocol proceeds in six steps: equilibrate reagents and TMB substrate to room temperature and set up standards, samples and control (zero) wells on the pre-coated plate; incubate the plate at 37°C for 90 minutes for antigen binding; add biotin-labeled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; develop colour with TMB substrate in the dark for 10–20 minutes; then add stop solution and read absorbance at 450 nm. Kit components are intended to be stored at 2-8°C for 12 months.

Reported linearity (n=5) at 1:2, 1:4 and 1:8 dilutions is 88-102%, 83-97% and 88-100% for serum, 98-104%, 85-101% and 84-97% for EDTA plasma, and 88-101%, 83-100% and 83-98% for heparin plasma. Recovery ranges and averages are 87-96% (average 92) for serum, 92-103% (average 97) for EDTA plasma, and 90-103% (average 98) for heparin plasma.
